Resveratrol-- Mechanism of action of resveratrol
Michael Derrida
The previous chapters have illustrated the diverse protective abilities of the n ovel phyto-pharmaceutical known as resveratrol against a broad spectrum of degen e rative diseases and dysfunctions. The following are major mechanistic pathways o f cytoprotection by this novel phytoalexin. Resveratrol:
*inhibits hydroxyl and peroxyl radicals
*induces Phase Il-drug-metabolizing enzymes and potentiates cancer anti-initiati on activity
*mediates anti-inflammatory effects and inhibits cyclooxygenase (COX) and hydrop eroxidase functions
*induces cancer antipromotion activity and potentiates cancer antiprogression ac tivity
*induces human promyelocytic leukemia cell differentiation
*induces carcinogen detoxification by inducing quaninone reductase activity
*inhibits ribonucleotide reductase, cyclooxygenase, cytochrome P450 1A1, DNA pol ymerase, and NADH:ubiquinone oxireductase enzyme activities. This ability to inh i bit these key enzymes explains resveratrol's antimutagenic, antiproliferative, a n d anti-carcinogenic potential.
*regulates nuclear transcription factor, NF-??, at the level of nuclear transloc ation
*inhibits brain monoamine oxidase in vivo
*enhances collagen synthesis
*functions as a novel phytoestrogen
*inhibits the expression of adhesion molecules ICAM-1 and VCAM-I.
